Tumor-derived alpha-fetoprotein (tAFP) causes immune and metabolic dysfunction in monocyte-derived dendritic cells

چکیده
Background Alpha-fetoprotein (AFP) is an oncofetal antigen expressed by over 50% of hepatocellular carcinoma (HCC) tumors. AFP-L3 is the major isoform present in the serum of HCC patients and is associated with poor patient prognosis. While HCC tumor-derived AFP (tAFP) contains >80% of AFP-L3, cord blood serum-derived AFP (nAFP) contains less than 5% of AFP-L3. Previous studies have proposed an immunoregulatory role for AFP on myeloid cells including dendritic cells (DC).

Tumor-derived alpha-fetoprotein impairs the differentiation and T cell stimulatory activity of human dendritic cells
Several tumor-derived factors have been implicated in DC dysfunction in cancer patients. Alpha-fetoprotein (AFP) is an oncofetal antigen that is highly expressed in abnormalities of prenatal development and several epithelial cancers, including hepatocellular carcinoma (HCC). In HCC patients exhibiting high levels of serum AFP, we have observed a lower ratio of myeloid-toplasmacytoid circulatin...
متن کاملTumor-derived alpha fetoprotein directly impacts human natural killer cell activity and viability
Alpha-fetoprotein (AFP) is an oncofetal antigen produced by hepatocellular carcinomas (HCC). Previous studies demonstrated that tumor-derived AFP (tAFP) is a glycoprotein that has an immunosuppressive role on natural killer (NK), T, B, and dendritic (DC) cells which may play a role in HCC pathogenesis. Defects in NK cells have been attributed to tAFP-mediated immunosuppression of DC. However, a...

متن کاملEvaluating the Effects of Cytomegalovirus Glycoprotein B on the Maturation and Function of Monocyte-derived dendritic cells
Background & Objectives: Interaction of cytomegalovirus glycoprotein B with toll-like receptors of dendritic cells leads to early signaling and innate immune responses. The aim of this study is to evaluate the effects of cytomegalovirus glycoprotein B on the maturation and function of monocyte-derived dendritic cells in treated groups in comparison with control groups. Materials & Methods: Bloo...
متن کاملDIFFERENTIATION OF MONOCYTE DERIVED DENDRITIC CELLS IN SERUM FREE CONDITIONS
Human peripheral blood monocytes (HPBM) were cultured in the absence of human serum and were converted into a state exhibiting a high accessory function expressed by their ability of supporting lymphocyte proliferation. After a prolonged culture in serum free media the monocyte derived cells were highly viable, increased in size and developed veils and dendritiform elongatio'l1s. Paralleli...
